大鼠骨髓间充质干细胞向血管平滑肌细胞分化过程中小窝蛋白-1表达增强

Expression of caveolin-1 is enhanced in the differentiation of rat bone marrow mesenchymal stem cells into vascular smooth muscle cells

摘要: 目的 探讨大鼠骨髓间充质干细胞向血管平滑肌细胞分化过程中小窝蛋白-1(caveolin-1)的表达变化。方法 采用全骨髓培养法体外分离培养SD大鼠骨髓间充质干细胞,成骨成脂成软骨检测干细胞的分化潜能,流式细胞仪鉴定干细胞表面标志物。体外TGF-β1诱导骨髓间充质干细胞向血管平滑肌细胞分化,Western blot检测SM-MHC、SM22α、caveolin-1和myocardin的表达。结果 骨髓间充质干细胞在TGF-β1诱导下可分化为血管平滑肌细胞,诱导培养组细胞caveolin-1、 SM-MHC及myocardin表达的水平随诱导时间的增加而增强(P<0.05)。结论 骨髓间充质干细胞体外诱导可以分化为血管平滑肌细胞, 在该分化过程中小窝蛋白-1的表达增强,提示小窝蛋白-1与干细胞来源的血管平滑肌细胞的形成有关。

Abstract: Objective To explore the expression of caveolin-1 in the differentiation of rat bone marrow mesenchymal stem cells(BMSCs) into vascular smooth muscle cells(VSMC). Methods BMSCs from SD rats were cultured in vitro by whole-bone marrow culture. Then, testing the differentiation potential of stem cells through adipogenic, osteoplastic and chondrogenic differentiation. BMSCs were induced to differentiate into VSMC by transforming growth factor-β 1(TGF-β1) in vitro. Western blot was performed to evaluate the expression of SM-MHC, SM22α and caveolin-1. Results TGF-β1 treatment increased the expression of caveolin-1, SM-MHC and myocardin in a time dependent manner. During the stage of BMSCs differentiate into vascular smooth muscle cells, the expression of caveolin-1 was specifically increased. Conclusions This study implies that caveolin-1 may be related to the formation of smooth muscle cells derived from stem cells.
