盐酸多奈哌齐增加血管性痴呆大鼠海马TGF-β和βFGF表达

Donepezil hydrochloride increases the expression of TGF-β and βFGF in the hippocampal neurons of mice with vascular dementia

摘要: 摘 要: 目的 研究盐酸多奈哌齐对血管性痴呆 (VD) 大鼠海马转化生长因子β(TGF-β)和碱性成纤维细胞因子(βFGF)表达的影响,探讨其对海马保护作用的机制。方法 120只雄性SD大鼠经Morris水迷宫筛选后随机分为假手术组、血管性痴呆模型组(模型组)、盐酸多奈哌齐药物干预组(药物组)。采用永久性结扎双侧颈总动脉法(2-VO)制作血管性痴呆动物模型。免疫组化法检测TGF-β、βFGF,TUNEL法检测凋亡细胞的表达变化。结果 与假手术组相比,模型组和药物组海马区TGF-β和βFGF表达增高,药物组海马区TGF-β(128.45±6.23)和βFGF(80.95±9.48)表达明显高于模型组(142.93±10.22;110.45±9.75。p<0.05)。假手术组和药物组细胞凋亡少,模型组细胞凋亡明显增多。结论 盐酸多奈哌齐可显著改善VD大鼠的学习和认知功能,增加TGF-β和βFGF表达,减轻神经元凋亡,保护神经细胞。

Abstract: Abstract: Objective To study the effects of Donepezil Hydrochloride on the expression of TGF-β、βFGF in the Hippocampal Neurons of Mice with Vascular Dementia. Methods 120 SD mice were divided into shame group(Shame),model group(Model) and treatment group with donepezil hydrochloride (Drug)randomly after the water maze test .The rat model of the vascular dementia was made by a Permanent bilateral occlusion of both common carotids arteries using 2-VO method. The expression of TGF-β and βFGF in hippocampus CA1 were observed by immunohistochemistry technique. The expression of apoptosis neuron were observed by TUNEL technique. Results The expression of TGF-β(128.45±6.23), βFGF(80.95±9.48) in hippocampus CA1 was significantly higher in the drug group than that in the model group(142.93±10.22;110.45±9.75.p<0.05).The expression of apoptosis neuron was low in the shame group and the drug group, but high in the model group. Conclusion Donepezil hydrochloride can increase the expression of TGF-β and βFGF, reduce the apoptosis neuron, and furthermore improve the abilities of learning and memory.
