参麦注射液促进心肌梗死模型大鼠心肌血管新生

Promoting effect of Shenmai injection on myocardial angiogenesis in rat models with myocardial infarction

摘要: 目的 研究参麦注射液对心肌梗死(MI)大鼠心肌血管新生的促进作用。方法 41只MI大鼠分为模型组、低和高剂量组,各10只,阳性组11只。低、高剂量组为1、2 mL/kg参麦注射液腹腔注射;阳性组为2 mg/kg依那普利灌胃。HE染色观察心肌组织病理变化;免疫组织化学、荧光染色观察梗死周围区毛细血管、小动脉密度;Western blot检测磷脂酰肌醇3-激酶(PI3K)、蛋白激酶B(AKT)、p-AKT、血管内皮生长因子(VEGF)蛋白表达量。结果 低和高剂量组、阳性组的病理改变减轻。与假手术组比较,模型组毛细血管、小动脉密度均增加(P<0.05),PI3K、VEGF蛋白表达量、p-AKT/AKT均升高(P<0.05);与模型组比较,低和高剂量组、阳性组的毛细血管、小动脉密度均增加(P<0.05),PI3K、VEGF蛋白表达量、p-AKT/AKT均升高(P<0.05)。结论 参麦注射液可促进MI大鼠心肌血管新生,推测与激活PI3K/AKT信号通路有关。

Abstract: Objective To study the promoting effect of Shenmai injection on myocardial angiogenesis in myocardial infarction(MI) rats. Methods Forty-one MI model rats were divided into model group, low and high-dose groups, with 10 rats each, and 11 rats in the positive group. Low and high-dose groups were treated with 1, 2 mol/kg of Shenmai injection injected intraperitoneally, positive group was treated with 2 mg/kg enalapril. HE staining was used to observe the pathological changes of myocardial tissue in each group. Immunohistochemical staining was used to observe the capillary density in the infarct area. Western blot was used to detect the relative expression of PI3K, AKT, p-AKT and VEGF protein. Results The pathological changes of the low and high-dose groups and the positive group were reduced. Compared with the sham operation group, the capillary density and arteriole density, the expression of PI3K and VEGF protein, and p-AKT/AKT in the model group were increased (P<0.05). Compared with the model group, the capillary density and arteriole density, the expression of PI3K and VEGF protein, and p-AKT/AKT in the low and high-dose groups and the positive group were increased (P<0.05). Conclusions Shenmai injection may promote myocardial angiogenesis in MI rats in a dose-dependent manner. It is speculated that the mechanism of this drug is related to the activation of PI3K/AKT signaling pathway.
