性别差异对苯肼致溶血性贫血小鼠的影响

Sex differences found in model mice of phenylhydrazine-induced anemia

摘要: 目的 观察性别因素对苯肼(PHZ)致溶血性贫血(HA)小鼠的影响。方法 10~11周龄C57BL/6小鼠经腹腔注射PHZ 溶液(120 mg/kg),于注射前和注射后第3、5、7、9和14天采颌下静脉血,经全自动血细胞分析仪检测红细胞(RBC)和白细胞(WBC)相关指标的变化。结果 注射PHZ后第3天,雌性和雄性小鼠均表现出相同程度的急性HA症状:皮肤黏膜颜色苍白、出现血红蛋白尿、体质量下降、RBC数量和红细胞压积(HCT)均显著性降低(P<0.05);注射PHZ后第3天,雌性小鼠脾脏代偿性增大的程度显著高于雄性(P<0.05);注射PHZ后第9天,雌性小鼠的RBC数量、HCT和血红蛋白含量(HGB)均显著高于雄性(P<0.05)。结论 PHZ引发的应激性红细胞生成过程存在性别差异:雌性小鼠RBC的回升速率大于雄性小鼠。

Abstract: Objective To observe the effect of gender on animal model in phenylhydrazine(PHZ)-induced hemolytic anemia(HA). Methods Ten to 11 week old C57BL/6 mice were injected intra-peritoneal with PHZ solution (120 mg/kg). Peripheral blood was collected from submandibular vein before PHZ injection and on the 3rd, 5th, 7th, 9th and 14th days after PHZ injection. The indexes of red blood cells and white blood cells were detected by auto hematology analyzer. Results On the third day after PHZ injection, both female and male mice showed similar symptoms of acute HA: pale paw skin; hemoglobinuria; weight loss; decreased erythrocyte counting and hematocrit(P<0.05). On the third day after PHZ injection, the compensatory increase of spleen size in female mice was significantly higher than that in male mice(P<0.05). On the 9th day after PHZ injection, erythrocytes number, hematocrit and hemoglobin content of female mice were also significantly higher than that of male mice(P<0.05). Conclusions There are sex differences in stress erythropoiesis induced by PHZ in mice. The recovery rate of erythropoiesis in female mice is higher than that in male mice.
