病毒拟似物Poly(I:C)诱导人卵巢颗粒细胞病毒反应蛋白viperin表达

摘要: 目的 探讨病毒拟似物对人卵巢颗粒细胞病毒反应蛋白表达的影响,明确颗粒细胞在卵巢病毒感染时的反应。 方法 将原代培养的人卵巢颗粒细胞分为对照组、病毒拟似物Poly(I:C)处理组、卵泡刺激素(FSH)处理组。 CCK8法测定细胞增殖活力、real-time PCR法测定干扰素-?(IFN-?)、viperin mRNA表达,Western blot法测定viperin蛋白表达。 结果 原代接种培养5 d后,FSH可诱导颗粒细胞增殖(P < 0.05),Poly(I:C)10 μg/mL对细胞增殖活力无显著影响。 Poly(I:C)刺激后24 h内诱导IFN-?、viperin mRNA表达明显上调,其中IFN-?于刺激后2 h表达水平达到峰值(P < 0.05),viperin于刺激后8 h达到峰值(P < 0.05),刺激后24 h,两者mRNA表达水平均回归刺激前水平。Poly(I:C)诱导颗粒细胞viperin蛋白表达,水平随Poly(I:C)浓度上升。结论 病毒拟似物能够诱导颗粒细胞表达病毒反应蛋白viperin。

Abstract: Objective To investigate the effect of Poly(I:C) on the expression of a virus responsive protein, viperin. Methods Human granulosa cells were obtained from patients who underwent in vitro fertilization. After incubation with FSH or Poly(I:C), the proliferation of granulosa cells were detected by CCK8 method. The expression of viperin, IFN-? were examined by real time PCR after a certain period of time after Poly(I:C) stimulation within 24 hs. The protein expression of viperin were examined by Western blot analysis. Results Ten μg/mL of Poly(I:C) has no significant influence on GCs’ proliferation. Poly(I:C) significantly induced mRNA expression of IFN-? (2 to 4 hs after stimulation) and viperin (8 to 12 hs after stimulation) (P < 0.05). Viperin expression was induced by Poly(I:C) in a dose-dependent manner. Conclusions Poly(I:C) induced IFN-? and viperin expression in human granulosa cells, which indicate that granulosa cells play an active role in anti-virus response upon infection.
