S100B、AQP4和CX43表达紊乱对糖尿病并发抑郁症大鼠海马血脑屏障功能的影响

Effects of S100B, AQP4, and CX43 expression on the function of the blood brain barrier in hippocampus of diabetic rats with depression

摘要: 目的 探讨S100B、AQP4和CX43表达紊乱对糖尿病并发抑郁症大鼠海马血脑屏障功能的影响。方法 将大鼠分为糖尿病组(高脂乳灌胃2周+尾静脉注射链脲佐菌素(STZ,38mg/kg))、抑郁症组(慢性不可预知性应激28 d)、糖尿病并发抑郁症组(联合以上两种方法建立)、对照组。造模结束后,Open-field和Morris水迷宫实验观察大行为学,ELISA检测血清S100B含量,免疫组化法检测海马血脑屏障AQP4和CX43的表达。结果 与对照组比较,糖尿病组和抑郁症组大鼠自主活动次数和空间探索时间显著减少,逃避潜伏期延长(P<0.05 or P<0.01);血清S100B含量明显增多(P<0.01);海马血脑屏障功能性蛋白AQP4、CX43积分吸光度明显减少(P<0.01)。与糖尿病组比较,糖尿病并发抑郁症组大鼠自主活动次数和空间探索时间减少,逃避潜伏期延长(P<0.05 or P<0.01);血清S100B含量明显增多(P<0.05);海马血脑屏障功能性蛋白AQP4、CX43积分吸光度明显减少(P<0.05 or P<0.01)。结论 S100B、AQP4和CX43表达紊乱所致海马血脑屏障功能障碍可能是糖尿病并发抑郁症发生机制之一。

Abstract: Objective To investigate.the effect of S100B, AQP4, and CX43 on the function of the blood brain barrier in the hippocampus of diabetic rats with depression.Methods Rats were divided into the diabetic group(tow weeks of high-fat diet and injection of Streptozotocin (STZ, 38 mg/kg)), the depression group(chronic unpredictable stress for 28 days), the diabetes mellitus with depression group(combined with the above two methods to establish), and the control group.The behavior of rats were evaluated with Open-field test and Morris test. The expression of AQP4 and CX43 in rat hippocampul blood-brain barrier was detected by immunocytochemistry. S100B was detected by ELISA . Results Compared with control group , the number of autonomic activity and was the space exploration times were decreased, the escape latencies were significantly longer in the Morris water maze test(P<0.05 or P<0.01) of the diabetes group and depression group; Serum S100B levels increased significantly(P<0.01);the expression of AQP4 and CX43 was decreased(P<0.01). Conclusions S100B, AQP4, CX43 expression disorder may be one of the mechanisms of diabetes mellitus complicated with depression.
