摘要: 肿瘤细胞不同于正常细胞,即使在氧气充足的条件下,主要依赖糖酵解代谢,称为有氧糖酵解。丙酮酸激酶(PK)是糖酵解最后一个限速酶,催化磷酸烯醇式丙酮酸产生丙酮酸和ATP。M2型丙酮酸激酶(PKM2),丙酮酸激酶同工酶之一,在肿瘤细胞中高表达。PKM2不仅在肿瘤代谢中,而且在基因表达的调控及细胞增殖中都具有重要作用。

Abstract: Unlike normal cells, cancer cells can generate energy through rapid glycolysis even in the presence of ample oxygen,which is known as aerobic glycolysis. Pyruvate kinase (PK) regulates the final rate-limiting step of glycolysis and catalyzes the dephosphorylation of phosphoenolpyruvate (PEP) to produce pyruvate and ATP. Pyruvate kinase M2 (PKM2), one of the PK isoenzymes, is specially expressed in cancer cells. PKM2 play a central role not only in metabolic reprogramming but also in direct regulation of gene expression and subsequent cell cycle progression.
