一种改良的蛋白质电泳考马斯亮蓝G-250染色方法

An improved method for protein staining with CBBG-250 following electrophoresis

摘要: 目的 建立一种简便、低毒的蛋白质聚丙烯酰胺凝胶电泳染色方法。方法 电泳后的凝胶经短暂固定后,用含氨基磺酸的考马斯亮蓝G-250溶液染色,无需脱色。结果 该方法灵敏度高于经典考马斯亮蓝R-250染色方法,并去除了常规染色和脱色液中有毒的甲醇等溶剂。结论 改良方法具有灵敏度高、染色背景浅、节省染料、直接显色无需脱色、配方安全低毒等优点。

Abstract: Objective: To establish a simple and less toxic method for protein staining following SDS-PAGE. Methods: After electrophoresis, the gel was fixed temporarily, and then stained with the Coomassie Brilliant Blue G-250 (CBBG-250) solution containing aminosulfonic acid. De-staining was not required. Results: The sensitivity of this method was higher than that of the classical CBBR-250 staining method. Moreover, toxic solvents such as methanol, though commonly used in the staining and de-staining solutions, were eliminated. Conclusion: The improved method has a series of advantages including high sensitivity, clear background, dye-saving, direct color development without de-staining, and having a safe and less toxic recipe.
