白血病患者血浆触珠蛋白-β链糖链唾液酸化与预后关系初探

Probing the relationship between sialylated glycopeptides from haptoglobin-beta chain in plasma and prognosis of patients with leukemia

摘要: 目的 探究白血病患者预后与血浆触珠蛋白-β链糖链唾液酸化之间的关系。方法 富集分离正常人及白血病患者不同治疗时间点血浆中触珠蛋白-β链,胶内胰酶及蛋白酶K双酶切,利用液质联用技术在线富集分离糖肽,以m/z 1045(3+)及m/z 948(2+)的唾液酸化糖肽相对峰强度定量分析唾液酸化程度,分析糖链丰度的变化。结果 白血病患者血浆触珠蛋白-β链糖链唾液酸化程度远高于正常人水平,重症患者尤为明显;在治疗好转后,唾液酸化水平逐渐降低接近于正常水平。结论 触珠蛋白-β链的变化趋势及其糖链唾液酸化程度的变化能反映白血病患者治疗过程中机体状态、疾病转归及预后。

Abstract: Object To analyze the relationship between sialylated glycopeptides from haptoglobin-beta chain in plasma at different treat time points and prognosis of patients with leukemia. Methods Plasma glycoproteins collected different treat time points, of the eight patients were enriched using lectin and then haptoglobin-beta chain were separated by gel. Resulting proteins were digested using trypsin, followed by proteinase K in gel, and the glycopeptides were enriched using liquid chromatography, followed by the structural characterization using tandem mass spectrometry. The change trends of sialylation were also analyzed. Results A decrease in the intensity of the sialylated glycopeptides of haptoglobin-beta chain was observed with disease recovery. Conclusion The changes in the level of the sialylated glycopeptides of haptoglobin were related to the prognosis of patients with leukemia. These sialylated glycopeptides may be a novel marker for monitoring the prognosis of leukemia.
