S-腺苷甲硫氨酸改善OLETF大鼠胰岛素敏感性

S-adenosyl-L-methionine increase insulin sensitivity in OLETF rats

摘要: 目的 观察S-腺苷甲硫氨酸对肥胖的2型糖尿病模型OLETF鼠骨骼肌mtDNA量和胰岛素敏感性的影响。方法 每日给大鼠腹腔内注射S-腺苷甲硫氨酸(15 mg/ kg )或安慰剂,测定食物摄入量、体质量及骨骼肌mtDNA量。行静脉葡萄糖耐量试验和正葡萄糖高胰岛素钳夹实验评价胰岛素敏感性。结果 S-腺苷甲硫氨酸治疗后OLETF 大鼠进食量减少,体质量增加幅度下降,骨骼肌mtDNA 量增加,葡萄糖输注速度增加,骨骼肌mtDNA 量与体质量呈负相关(?=0.696,p<0.05)。结论 S-腺苷甲硫氨酸可以改善OLETF鼠胰岛素敏感性,抑制体重增加,增加骨骼肌mtDNA量。

Abstract: Objective To investigate the effective of S-adenosyl-L-methionine (SAMe) on skeletal muscle mitochondrial DNA density and insulin sensitivity in Otsuka Long-Evans Tokushima Fatty (OLETF) rats. Methods SAMe (15 mg /kg?d ) or placebo was administered intraperitoneally for 6-week-old OLETF rats. Food intake, body weight and skeletal muscle mitochondrial DNA density at different age were determined. Insulin sensitivity was evaluated by euglycemic-hyperinsulinemic clamp test. Results SAMe treatment significantly inhibited food intake and reduced body weight gain as well as increased skeletal muscle mitochondrial DNA density and whole body insulin sensitivity in OLETF rats compared with their controls. Furthermore, skeletal muscle mitochondrial DNA density correlated with insulin body weight (?=0.696. p<0.05). Conclusion SAMe treatment increased mitochondrial DNA density in the skeletal muscle , prevented body weight gain, and improved whole body insulin sensitivity in OLETF rats.
