高浓度氧增加未成年大鼠肺血红素加氧酶-1表达

Hyperoxia increases the expression of lung heme oxygenase-1 in rat pups

摘要: 目的 探讨高浓度氧对未成年大鼠肺血红素加氧酶-1(HO-1)表达的影响。方法 将生后 21 d SD 大鼠 40 只随机分为空气组和 12、24、48 及72 h 高氧组,分别置于空气和常压高氧箱(92%~94% O2)中。检测左肺湿/干重比及肺组织病理学改变,用 RT-PCR 和 Western blot 法分别检测肺 HO-1mRNA 及 HO-1 蛋白表达情况。结果 高氧 12 h 组肺 HO-1 mRNA 吸光度积分相对值和高氧 24 h 组 HO-1 蛋白表达水平分别为 0.35 ± 0.043 和 0.455 ± 0.046,较对照组0.263 ± 0.037 和 0.28 ± 0.044 明显升高,且均随高氧暴露时间延长而表达进一步增加(P < 0.05,P < 0.01)。与空气组比较,高氧 48 h 组和高氧 72 h 组左肺湿重/干重及肺损伤评分显著增高(P < 0.05,P < 0.01)。结论 高浓度氧可引起未成年大鼠肺组织 HO-1 表达增多。

Abstract: Objective To investigate the effects of hyperoxia on the expression of lung heme oxygenase-1 (HO-1) in rat pups. Methods 21-d-old male Sprague-Dawley rat pups were randomly divided into 5 groups: room-air control group and 12, 24, 48, 72 hours hyperoxia groups. The rat pups were continuously exposed to oxygen (92%~94%) and room-air respectively. The left lung wet/ dry weight ratio and the histological examination were performed. The expressions of lung HO-1 mRNA and HO-1 protein were also measured by RT-PCR and Western blot, respectively. Results The expressions of lung HO-1 mRNA in 12 hours hyperoxia group (0.35±0.043) and HO-1 protein in 24 hours hyperoxia group (0.455±0.046) were increased as compared to those in the room-air group (0.263±0.037, 0.28±0.044), and the increases were more significant with the longer hyperoxia exposure (P < 0.05, P < 0.01). Compared with the room-air control, the left lung wet/ dry weight ratio and the lung injury scores were significantly increased in 48 hours hyperoxia group and 72 hours hyperoxia group (P < 0.05, P < 0.01). Conclusion Hyperoxia can induce the increase of lung heme oxygenase-1 expression in rat pups.
