大鼠心肌微血管内皮细胞的体外原代培养

摘要: 目的 体外原代培养大鼠心肌微血管内皮细胞(MMVECs),并观察其生长特性。方法采用酶蛋白消化法培养大鼠MMVECs,用差速贴壁法和亚细胞克隆法纯化细胞,用第Ⅷ因子相关抗原和CD31相关抗原鉴定细胞,倒置相差显微镜下观察细胞生长状态。结果 成功培养出纯度很高的MMVECs,可用于MMVECs相关实验研究。结论 此原代培养MMVECs的方案具有较好的可操作性和可重复性。

Abstract: Objective To improve methods of primary culturing rats myocadial microvascular endothelial cells in vitro,and observing the growth feature. Methods Enzyme digestion cultivation of cadiac tissue was used to obtain the primary cells.In the subculturing, the cell cultures were purified by different speeds of attachment and sub-cell cloning method. The purified cells were identified as endothelial cells by factor Ⅷ and CD31 related antigen ,the two markers for endothelial cells,and observing the growth feature by inverted phase contrast microscope. Results The MMVECs were cultured and purified successfully which can be used in the related researches. Conclusion  The protocol of culturing MMVECs was succeeded, and it is reliable.
