曲妥珠单抗降低乳腺癌细胞条件培养液促血管内皮细胞增殖

Trastuzumab decreases the proliferating role on endothelium induced by TCM from MDA-MB-231

摘要: 目的 观察表皮生长因子受体-2(HER-2)的特异阻断剂曲妥珠单抗对乳腺癌细胞条件培养液促内皮细胞增殖、抗凋亡效应的影响。方法 培养并收集乳腺癌细胞MDA-MB-231条件培养液作用于内皮细胞,并用曲妥珠单抗进行干预,然后用MTT、流式细胞仪检测细胞的增殖和凋亡;免疫组化检测乳腺癌组织微血管密度与HER-2表达。 结果 MDA-MB-231的条件培养液可显著促进脐静脉内皮细胞(HUVEC)的增殖和抗凋亡(P<0.05);曲妥珠单抗显著减弱上述作用(P<0.05),但对内皮细胞的增殖、凋亡无直接影响。35例乳腺浸润性导管癌病理中,19例HER-2(+),16例HER-2(-)。微血管的面积密度分别为(0.72±0.77,0.74±0.11)和数密度分别为(33.83±3.89 /mm2,36.42±5.65 /mm2),两组之间均无差异。结论 曲妥珠单抗可显著降低乳腺癌细胞条件培养液的促内皮细胞增殖和抗凋亡效应。

Abstract: Objective To investigate the effect of trastuzumab, a special blocker of epidermal growth factor receptor-2(HER-2) on proliferation and apoptosis of human umbilical vein endothelial cells (HUVECs) exposed to tumor conditioned medium(TCM). Methods HUVECs were cultured and exposed to TCM, and trastuzumab was used as an intervention. Then their proliferation, apoptosis and cell cycle distribution were measured by MTT and flow cytometery(FCM). Intra-tumor microvessel density (MVD) and HER-2 expression were tested by immunohistochemistry(IHC). Results Following the stimulation of MDA-CM, HUVECs showed higher proliferating and anti-apoptotic ability(P<0.05). Trastuzumab relieved the above roles significantly((P<0.05), while trastuzumab didn't affect HUVEC's growth directly. Among 35 cases of breast invasive ductal carcinoma, 19 cases were HER-2 negative with an area density of MVD (0.72±0.77 and count density(33.83±0.11/mm2), 16 cases were positive with a relative higher value( 0.74±0.11 and 36.42±5.65/mm2 respectively), the discrepancy has no significance. Conclusion Trastuzumab could decrease the proliferating and anti-apoptotic role on endothelium induced by TCM from MDA-MB-231.
