Sphingosine 1-phoshate prolongs action potential duration and inhibiting voltage-dependent potassium current in guinea-pig ventricular myocytes

摘要: 目的 研究1-磷酸鞘氨醇(S1P)对豚鼠心室肌细胞动作电位(AP)及电压依赖性钾电流(KV)的作用及其机制。方法 利用Langendorff离体心脏逆向灌流法分离豚鼠心室肌细胞,采用全细胞膜片钳技术记录心室肌细胞AP和KV。结果 S1P可延长动作电位复极50%时程(APD50)和动作电位复极90%时程(APD90),降低KV,其作用可被百日咳毒素(PTX)和卡弗他丁C(Calphostin C)阻断。结论 S1P可能通过PKC途径抑制KV及延长AP时程。

Abstract: Objective To investigate the effect and mechanism of sphingosine 1-phoshate (S1P) on action potential (AP) and voltage-dependent potassium current (KV)in isolated guinea pig ventricular myocytes. Methods The ventricular myocytes were isolated by using Langendorff perfusion method. The AP and KV were recorded by whole cell patch-clamp recording technique. Results S1P prolonged the 50% and 90% action potential duration (APD50 and APD90) and decreased KV which were blocked by pertussis toxin (PTX) or Calphostin C. Conclusion S1P decreased the KV in a PKC pathway and prolonged action potential duration in guinea-pig ventricular myocytes.
