PTEN与MGMT蛋白在男性乳房发育症中的表达

Expression of PTEN and MGMT protein in gynecomastia

摘要: 目的 观察抑癌基因PTEN和DNA直接修复酶MGMT在男性乳房发育症(GM)乳腺中的表达。方法 采用免疫组化SP法检测68例GM乳腺(实验组)和24例正常男性乳腺(对照组)中PTEN与MGMT的表达状况。将所选实验组分别按年龄及组织学类型分组并分别研究。结果 PTEN与MGMT表达均位于乳腺导管上皮细胞核。GM乳腺中PTEN与MGMT表达水平较正常男性乳腺 显著降低(P<0.01)。二者在GM不同年龄组间的表达有差异(P<0.05),且PTEN表达水平在中年组、老年组、青年组依次降低;MGMT表达水平在老年组、中年组、青年组依次降低。二者在GM不同组织学类型间的表达亦有差异(P<0.05),且二者表达水平在硬化型、过渡型、旺炽型均依次降低。PTEN与MGMT表达之间没有相关性。结论 GM乳腺组织中PTEN与MGMT表达的异常可能与GM的发生有关。

Abstract: Objective To detect the expression of a new tumor suppressor gene PTEN and DNA direct repair enzyme MGMT in gynecomastia. Methods Immunohistochemical SP method was used to detect expression of PTEN and MGMT protein in 68 cases of gynecomastia(experiment group) and 24 cases of mammary gland of normal man tissues(control group). The selected examples were divided into three different age groups and three different histological types. Results The PTEN and MGMT protein were all expressed in nuclear of ductal cellula epithelialis. The expression level of PTEN and MGMT protein in gynecomastia was significantly lower than mammary gland of normal man tissues(P<0.01). There was difference between the expression of PTEN and MGMT protein in different age groups(P<0.05), moreover, the expression level of PTEN protein degraded in middle-aged group, older age-group, and young group by turns;the expression level of MGMT protein degraded in older age-group, middle-aged group, and young group by turns. The expression of PTEN and MGMT protein in three different histological types was different(P<0.05), and the expression level of them degraded in constrictive type, transitional type, and prosperous and intense type by turns. Conclusion This study suggests that the abnormal expression of tumor suppressor gene PTEN and MGMT in gynecomastia probably plays an important role in the development of gynecomastia.
