NGF及其受体P75在四氯化碳中毒性大鼠肝组织中表达增强

Increased expressions of NGF and its receptor P75 in liver of CCl4-induced toxic rats

摘要: 目的 观察四氯化碳中毒时神经生长因子(NGF)及其受体P75在大鼠肝组织的表达变化,探讨NGF在肝纤维化发生机制中的可能作用。 方法 用四氯化碳致大鼠肝中毒模型,用RT-PCR法和Western blot法,分别检测NGF及其受体P75在大鼠肝组织中的表达。 结果 对照组大鼠肝组织中NGFmRNA及其受体P75蛋白表达量比较,造模组24h显著增高(P<0.01,n=6),造模48h 和72h迅速回降,但仍显著高于对照组(P<0.05,n=6)。 结论 在四氯化碳中毒性大鼠的肝组织中,NGF及其受体P75的表达可能与肝纤维化的形成密切相关。

Abstract: Objective By observing the changes in NGF and its receptor P75 in liver of CCl4-induced toxic rats,to evaluate the role of NGF in the mechanism of hepatic fibrosis. Methods The expressions of NGF mRNA and its receptor P75 were detecteded by both hemi-quantitative RT-PCR and Western blot analysis in liver of CCl4-induced toxic rats at the time of 24h,48h,72h,96h、1w、5w and 9w . Results The expressions of NGFmRNA and its receptor P75 in liver of CCl4-induced toxic rats at 24th hour group were higher than that of control group (P<0.01,n=6). Then it was swith low at 48th hour and 72th hour. But it was still higher than that of control group(P<0.05,n=6). Conclusion In CCl4-induced rat liver tissue,the expressions of NGF and its receptor P75 might be significantly related to hepatic fibrosis.
