基因枪转导K-ras反义基因对胰腺癌细胞K-ras p21蛋白表达的影响

Effect of gene gun transduction of Kras Antisense gene on the expression of Kras p21 protein in human pancreatic carcinoma cells

摘要: 目的:探讨基因枪转导K-ras反义基因对胰腺癌细胞K-ras p21蛋白的影响。方法:利用基因枪技术,将反义基因转导入宿主细胞,通过免疫细胞化学和Western Blot方法,观察K-ras突变特异性反义基因转导后AsPC-1、MiaPaCa-2和BxPC-3三种人胰腺癌细胞系K-ras p21蛋白的变化情况。结果:转导K-ras突变特异性反义基因后,突变型AsPC-1和MiaPaCa-2胰腺癌细胞K-ras p21蛋白的表达明显降低。结论:突变特异性K-ras反义基因经有效转导后,可显著下调突变型胰腺癌细胞系AsPC-1和MiaPaCa-2的K-ras p21蛋白表达,突变特异性K-ras反义基因可以达到胰腺癌基因治疗的目的

Abstract: Objective To investigate the effect of gene gun transduction of Kras Antisense gene on the expression of Kras p21 protein in human pancreatic carcinoma cells.Methods Kras Antisense gene was transduced into pancreatic carcinoma cells by gene gun transduction, the expression of Kras p21 protein on BxPC-3、AsPC-1 and MiaPaCa-2 pancreatic carcinoma cells line were observed by western blot and immunocytochemistry staining. Result The expression of Kras p21 protein in AsPC-1 and MiaPaCa-2 pancreatic carcinoma cells was lower obviously after transduction of mutation specific Kras Antisense gene. Conclusion Gene gun transduction of Kras Antisense gene can significantly down-regulate the expression of Kras p21 protein in AsPC-1 and MiaPaCa-2 pancreatic carcinoma cells, which can be an effective gene trerapy method in treating pancreatic carcinoma.
