Effect of Alprostadil on Antithrombotic Function in Rabbit Endothelial Cells Injuried by Hypoxia/Reoxygenation

Abstract

OBJECTIVE To study the effect of alprostadil (PGE1 on antithrombotic function in rabbit endothelial cells injuried by hypoxia/reoxygenation(H/R.METHODS H/R injury model was established with the cultured endothelial cells from rabbits in vitro.The endothelial cells were divided into normal group, model group and treatment groups with different concentrations of PGE1(0.05, 0.20, 0.40 μg·mL-1. PGE1 was added to the liquid of H/R and adjusted to a final concentration mentioned above . Malondialdehyde (MDA content, the activity of superoxide dismutase (SOD, and the nitric oxide (NO content in the culture supernatants and the expression of von Willebrand factor (vWF in cells sampled from different group (normal, model and treatment groups were assayed.RESULTS Compared with normal group,MDA content in model group was increased, while the activity of SOD, NO content and the expression of VWF in model group were reduced(P<0.05. Compared with model group, the activity of SOD and NO content in treatment groups with different concentrations of PGE1were increased, while MDA contents in treatment groups were reduced, and the expression of vWF was enhanced(P<0.05. CONCLUSION PGE1 has protective effect on antithrombotic function in rabbit endothelial cells injuried by H/R.
