Abstract
OBJECTIVE To study the pharmacokinetics of doxapram injection in Uigur and Han healthy volunteers. METHODS Ten Uigur and 10 Han healthy volunteers were given a single dose of 50 mg doxapram injection intravenously. The plasma concentrations of doxapram were determined by HPLC. The pharmacokinetic parameters were calculated with DAS2.0 software. RESULTS The plasma concentration-time curves of doxapram were fitted to a two-compartment model. The main pharmacokinetic parameters in Uigur and Han healthy volunteers were as follows： t1/2α (0.49±0.42) and (0.33±0.24)h， t1/2β (3.65±2.07) and (3.87±2.17) h， Vc (0.788±0.523) and (0.354±0.197)L·kg-1， Vd(1.812±0.986) and (1.350±0.957) L·kg-1， CL (0.394±0.251) and (0.245±0.113)L·h-1·kg-1， AUC0-12 (2.320±0.700) and (3.513±1.260)mg·h·L-1， AUC0-∞ (2.682±0.801) and (4.061±1.445) mg·h·L-1， respectively. CONCLUSION There is significant difference in the pharmacokinetic parameters Vc， AUC0-12， AUC0-∞ between Uigur and Han healthy volunteers.
