Preliminary Study on Characteristics of in situ Pollen Germination and Self-Incompatibility of Cistanche deserticola

Abstract

OBJECTIVE To study the characteristics of in situ pollen germination of C. deserticola and to judge the type of self-incompatibility of C. deserticola. METHODS Fluorescence microscopy was used to observe the in situ pollen germination and pollen tube growth in the stylar canal of C. deserticola. The characteristics in different phases after cross-pollination and self-pollination were compared, and the type of self-incompatibility was evaluated by generated location of callose in the pollentube. RESULTS Fluorescence microscopy showed that the pollen germinated 40 min after pollination, and the tubes were equally long as the diameter of pollen grain. But only parts of the pollens from self-pollination germinated and remained on the stigma. 10-12 h after cross-pollination, the pollen tubes gathered to the stylar cannl and were 8-15 mm far from stigma. 48-72 h after self-pollination, the pollen tubes almost ceased growing in the junction of the stigma and stylar, which were only 2.8-5.6 mm long. At the end of the tubes, the long bright callose spots were germinated. CONCLUSION The population of C. deserticola possesses the characteristic of self-incompatibility, and the type of self-incompatibility should be gametophytic type. The pollens and stigma from the same flower muture togother and have no much separation. Self-pollination is apt to emerge but with no seed setting. It may be one of the reasons for low seed setting percentage of C. deserticola.
