Abstract
OBJECTIVE To establish a HPLC for the quantitative determination of saikosaponin b1 and b2 in Chai-huang Tablet. METHODS The HPLC method was performed on a Hypersil C18 column (4.6 mm×250 mm, 5 μm with MeOH - Water (75∶25 as the mobile phase. The flow rate was 1.0 mL·min-1. The UV detection wavelength was 254 nm, and the column temperature was set at 30 ℃. RESULTS The calibration curves showed good linear response ranged from 0.015 to 0.120 μg (r=0.999 5 for saikosaponin b1 and from 0.057 to 0.456 μg (r=0.999 8 for saikosaponin b2. The average recoveries were 97.2% (RSD=2.0% and 96.9% (RSD=1.6% respectively. CONCLUSION The method is simple, reliable and repeatable, and can be used for the quantitative determination of saikosaponin b1 and b2 in Chai-huang Tablet.
