Abstract
OBJECTIVE To investigate the effect of hydrochloric higenamine on cardiovascular system after the administration of β-receptor blocker and to measure the result of stress experiment in coronary artery stenosis model. METHODS ■Selective β1-receptor blocker metoprolol tartrate and non-selective β-receptor blocker propranolol were intravenous injected in ten Chinese miniature pigs, respectively. Heart rate (HR), systolic blood pressure(SBP) and diastolic blood pressure(DBP) were recorded after higenamine was introduced, and compared with those treated with higenamine along. Drug stress test and static myocardial SPECT imaging in model of chronic coronary artery stenosis were performed. RESULTS Mean HR of pigs was increased (40±17) beat·min-1 (P<0.001), mean DBP was decreased (20±16) mmHg (P=0.003 5), SBP showed no significant change (P>0.05) after intravenous administration of higenamine. Mean HR of pigs was also increased by higenamine after metoprolol and propranolol intervention (27±17) and (8±9) beat·min-1, respectively. Compared with baseline, change of HR was significant (P=0.000 3; P=0.022 1, respectively). The sensitivity for the detection of coronary artery disease by radionuclide myocardial perfusion imaging with higenamine was 90%. CONCLUSION This study demonstrated that active β-receptor blocker, at least partially, contributed to effect on cardiovascular system by hydrochloric higenamine, β-receptor blocker could partially block effect and myocardial perfusion imaging with higenamine had high sensitivity for detecting coronary artery stenosis and ischemic myocardium.
