Antioxidant Activities of Flavonoids from Xinjiang Glycyrrhiza glabra

Abstract

OBJECTIVE To prepare licorice general flavonoids(LGFs) and its purified constituent glabridin from Xinjing Glycyrrhiza glabra L,and to investigate their antioxidant properties.METHODS The routine method was applied for obtaining LGFs and glabridin.Two different in vitro oxidative systems: the enzymatic oxidative system MPO(myeloperoxidase) /H2O2/HOCl in human HL-60 cell line and the cytochrome P450/NADPH-reductase within hepatic rat microsomes were used for testing their antioxidant properties.The free radical scavenger Ginkgo biloba extract(EGb 761) was used as reference.Cellular and microsomal reactive oxygen species(ROS) formation was determined by the fluorescence of 2′,7′-dichlorofluorescein(DCF) originated from 2′,7′-dichlorofluorescein diacetate(DCFHDA) probe oxidation.RESULTS In both models,the preparied substances exhibited globally remarkable antioxidant properties and were classified by their ROS inhibition capacity as follows: EGb761≥GLB>LFs.CONCLUSION The present study demonstrates that licorice general flavonoids and its active principle glabridin have relatively high antioxidant activities in both oxidative models.These results could partially explain the pharmacological actions of licorice with antioxidant prophylactic activities.
