Abstract
OBJECTIVE To develop a simple and rapid high-performance capillary electrophoresis(HPCE) coupled with UV detection method for the determination of rutin and hyperin in Acanthopanax senticosus(Rupr.et Maxim) leaves simultaneously.METHODS The buffer was composed of 0.2 mol·L-1 NaH2PO4,0.1 mol·L-1 Borax and double distilled water(10∶7∶3,pH 8.8) solution.Capillary electrophoresis was performed using a 25 μm×45 cm fused silica capillary column.Separation voltage was 20 kV,detection wavelength was set at 254 nm,and the injection time was 30 s at a height of 10 cm.RESULTS The linear range of rutin was from 125~750 mg·L-1,its linear regression equation was Y=144 145ρ+423.87(r=0.999 9),and the average recovery was 99.38%,while the linear range of hyperin was from 200~1 200 mg·L-1,its linear regression equation was Y=216 836ρ-191.57(r=0.999 9),and the average recovery was 100.39%.CONCLUSION This method can be used for the quality control of the Acanthopanax senticosus(Rupr.et Maxim) leaves with good precision.The contents of rutin and hyperin are different in Acanthopanax senticosus(Rupr.et Maxim) from different places and different collection periods.
