Phytoestrogenic Effects of Six Active Components in Chinese Medicine

Abstract

OBJECTIVE To evaluate the estrogenic effects and their mechanisms of six active components in Chinese medicine through the test of their influence on MCF-7 proliferation and its cell cycle.METHODS The proliferation of MCF-7 influenced by ecdysterone,saffcomin A,tanshinone I,tanshinone ⅡA,psoralen and isopsoralen was analyzed by MTT assay and their mechanisms were studied by adding ER antagonist ICI180,782.The cell cycle distribution of MCF-7 was determined by flow cytometry.RESULTS The proliferation rates of the cells treated with ecdysterone,saffcomin A,psoralen and isopsoralen were markedly increased.While the cell cycle was impulsed from G1 to S.DNA synthesis was inhanced and PI was increased.The above function on boosting proliferation was inhibited by adding estrogen receptor antagonism ICI182,780.Tanshinone I and tanshinone ⅡA showed inhibitory effects on proliferation of ER positive MCF-7 cell.However,no distinct effects were obtained on cell cycle.At the same time,the inhibitory effects of tanshinone I and tanshinone ⅡA were significantly decreased by adding the estrogen receptor antagonist ICI182,780.CONCLUSION Ecdysterone,saffcomin A,psoralen and isopsoralen can improve MCF-7 proliferation.While tanshinone I and tanshinone ⅡA have inhibitory effects on MCF-7 proliferation.All of their effects were realized through estrogen receptor.
