Abstract
OBJECTIVE To develop a optimized method for the determination of trans-resveratrol in rat urine. METHODS The sample was processed by using solid-phase extraction with polyamide cartridges. A central composite design, which allowed a sequential response surface methodology to he performed, was used for the optimization of the extraction procedure. RESULTS The optimized condition for SPE was achieved by using 10% methanol 10 mL,30% methanol 5 mL and 100% methanol 4 mL. The linear range was obtained from 0.043 to 8.6 mg·L-1 and the limit of detection was 0,043 mg·L-1. The average recovery was 98.0% and the RSDs of intra-day and inter-day were less than 10%.CONCLUSION A reliable and specific SPE-HPLC method for the determination of resveratol in rat urine was developed. This method effectively decreased the interference of endogenous compounds, and was appropriate for the determination of resveratrol in rat urine.
