Abstract
OBJECTIVE:To report a method for the determination of MS-551(MS) concentrations in plasma by HPLC.METHODS:MS was separated on a C18 column(5μm,4.6mm×15cm) by using methanol -0.1M ammonium acetate (60∶40) as mobile phase with detection wavelength at 270nm.Quantification was carried out based on the external standard of MS.RESULTS:There was a good linearity over the range from 0.1 to 2.0μg·ml-1 of MS,Y=8.6963×10-2+3.55×10-6X(r=0.9990).The average recovery was 94.27%.The detection limit was 0.85ng.At the same time,the relationship between the plasma concentrations of MS and QTc was determined in anaesthetized dogs with MS.CONCLUSION:This method is simple,practical and accurate.
