摘要
目的 建立高效液相法测定丹参中丹参素的含量,对不同地区、不同栽培方式的丹参进行含量比较,为制定其质量标准提供依据。方法 用Kromails C18(250 mm×4.6 mm,5 μm)色谱柱,流动相为甲醇-0.2%醋酸(10:90),检测波长为280 nm,流速为1.0 mL·min-1,温度为室温。结果 丹参素在0.252-1.26μm内呈良好线性关系,回归方程为y=7.13×105X-1.85×104,(r=0.9996),平均回收率为97.61%(RSD=1.37%,n=5)。结论 本法操作简便,结果准确,可作为丹参药材的含量测定方法,成为控制丹参药材质量的方法之一。应用本法,比较了不同地区、不同栽培方式的丹参,其中四川中江(栽培)产的丹参素含量最高,河北承德(栽培)产的含量最低。
Abstract
OBJECTIVE To establish the quality control standard for Salvia miltiorrhiza Bge.that was cultivated by different methods and in different regions. METHODS A RP-HPLC method was established for the determination of Danshensu in Salvia miltiorrhiza Bge.. Kromails C18(250 mmx4.6 mm, 5μm) column was used. The mobile phase was composed of methanol and 0.2% acetic acid (10:90) with the flow rate of 1.0 mL· min-1. Detection was taken at 280 nm at room temperature.The contents of Danshensu over different samples were determined. RESULTS The calibration curve of Danshensu was linear over the range of 0.252?1.26 μg. The regression equation was Y=7.13x105X-1.85X104(r=0.999 6).The average recovery was97.61% (RSD=1.37%, n=5). CONCLUSION The method was proved to be simple, accurate and could be used to control the quality of Salvia miltiorrhiza Bge.. The content of Danshensu was the highest in Salvia miltiorrhiza Bge.. cultivated in Zhongjiang city of Sichuan Province and the lowest in Chengde city of Hebei Province.
