摘要

目的 从山豆根中精制苦参碱和氧化苦参碱,并以此为化学对照品对山豆根药材进行薄层色谱鉴别。方法 利用硅胶柱色谱和重结晶的方法分离纯化苦参碱和氧化苦参碱,经理化数据和光谱解析等方法鉴定结构,采用TLC和高效液相色谱的方法检查其纯度,并利用TLC的方法对不同来源的山豆根商品药材进行鉴别研究。结果 从山豆根中分离纯化得到纯度98%以上的苦参碱和氧化苦参碱对照品各1.7 g和2.0 g,并以此为对照品有效地鉴别了正品山豆根药材及其混淆品。结论以苦参碱和氧化苦参碱作为山豆根的定性指标是合理的,采用TLC方法鉴别山豆根是可行的。

Abstract

OBJECTIVE To isolate and purify the two alkaloids matrine and oxymatrine and then to identify the commercial medicinal material of "Shandougen" by TLC. METHODS Matrine and oxymatrine were isolated from the roots of Sophora tonkinensis by repeated silica gel column chromatography and purified by recrystallization method. Their structures were elucidated on the basis of physical and chemical properties and spectral data, and the purities of these two compounds were determined by TLC and HPLC analysis. The crude drugs of "Shandougen" from different sources were identified by TLC using matrine and oxymatrine as the reference substances. RESULTS 1.7 g of matrine and 2.0 g of oxymatrine with the purity of more than 98% were prepared. The genuine samples of "Shandougen were differentiated from the adulterations by TLC method. CONCLUSION Matrine and oxymatrine are suitable to be used as the reference compounds for the TLC identification of "Shandougen".
