Protective Effect of Velvet Antler Peptides on Acute Hepatic Injury by Carbon Tetrachloride

OBJECTIVE To study the protective effects of velvet antler polypeptides(VAP) on acute hepatic injury induced by carbon tetrachloride(CCl4) in mice.METHODS Sixty Kunming mice were randomly divided into 6 groups.The VAP groups were treated with VAP at a dose of 12.5,25 and 50 mg·kg-1·d-1 for 5 days subcutaneously.At the same time,the normal control group and the model control group were given equivalent amount of 0.9% sodium chloride solution by the same route.And mice in the positive control group were intragastically administrated with 200 mg·kg-1·d-1 BDD.After the last administration,all groups except the normal control were intraperitoneally injected with 0.2% CCl4 solution to induce acute hepatic injury.Thereafter, the contents of ALT,AST,ALB in the serum,MDA and the activity of SOD,GSH in the liver were measured and the hepatic histological changes were observed by optical microscope.RESULTS Compared with the model group, the contents of ALT,AST in the serum was obviously decreased(P<0.01,P<0.001),the content of MDA in the liver was also content lowered(P<0.01),and the activity of GSH and SOD in the liver was markedly improved(P<0.05,P<0.01) after preventive treatment with VAP.VAP Ameliorated the hepatic pathological changes.CONCLUSION VAP have protective effects on acute hepatic injury induced by CCl4 in mice.
