摘要
目的 观察梅花鹿茸多肽(VAP)对四氯化碳所致小鼠急性肝损伤的保护作用。方法 腹腔注射四氯化碳(CCl4 ) 制备小鼠急性肝损伤模型,检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性和白蛋白(ALB)含量;同时测定肝组织中丙二醛 (MDA)含量和超氧化物歧化酶(SOD)、谷光甘肽(GSH)的活性。在光学显微镜下观察肝脏病理组织学变化。结果 与四氯化碳模型组比较,VAP可使血清ALT、AST活性明显降低(P<0.01),但对ALB的含量无明显影响(P>0.05);同时VAP可使肝组织中MDA的含量明显下降(P<0.01),GSH含量有所升高,SOD活力明显增强(P<0.01)。形态学观察,VAP能明显改善肝组织的病理学改变。结论 VAP对四氯化碳所致小鼠急性肝损伤具有保护作用。
Abstract
OBJECTIVE To study the protective effects of velvet antler polypeptides(VAP) on acute
hepatic injury induced by carbon tetrachloride (CCl4 ) in mice. METHODS Sixty Kunming mice were randomly divided into 6 equal groups. The VAP groups were treated with VAP at a dose of 12.5, 25 and 50mg•kg-1•d-1 for 5 days subcutaneously. At the same time, the normal control group and the model control group were given equivalent amount of 0. 9%sodium chloride solution by the same route . And mice of the positive control group were intragastically administrated with 200mg•kg-1•d-1 BDD. After the last administration, all groups except the normal control were intraperitoneally injected with 0.2% CCl4 solution to induce acute hepatic injury.Thereafter, the contents of ALT, AST, ALB, MDA and the activity of SOD, GSH were measured and the hepatic histological changes were observed by optical microscope. RESULTS As compared with the model group, the contents of ALT, AST obviously decreased (P<0.01, P<0.001) , the MDA content lowered (P<0.01), and the GSH content increased, the SOD activity improved ( P < 0.05, P<0.01) after preventive treatment with VAP. What’smore, VAP could ameliorate the hepatic pathological changes. CONCLUSION VAP have p- rotective effects on acute hepatic injury induced by CCl4 in mice.
