目的 建立指纹图谱与一测多评法同时测定积雪草中5种代表性三萜类成分的含量,并验证该方法在积雪草质量控制中的适用性与准确性。方法 首先采用HPLC建立积雪草药材指纹图谱;以积雪草为研究对象,以积雪草苷为内参物,建立积雪草酸、羟基积雪草酸、羟基积雪草苷、积雪草苷B与其之间的相对校正因子(fs/k),用fs/k计算其他4种成分的含量;同时对一测多评的计算值与外标法实测值进行比较。结果 以收集的12个批次积雪草样品为研究对象,建立了积雪草药材HPLC指纹图谱,确定了23个明显的共有峰,对8个共有峰进行了化学成分指认;建立了积雪草多成分一测多评定量方法,在各自线性范围内,积雪草苷B、羟基积雪草苷、羟基积雪草酸和积雪草酸的fs/k分别为0.85、1.13、0.46和0.45;且在不同高效液相色谱仪和不同色谱柱等条件下具有较好的重复性和耐用性;利用一测多评法与外标法得到的12批积雪草样品中各成分含量没有显著性差异。结论 建立的一测多评法可同时对积雪草中5种代表性三萜成分进行定量分析,该方法简便、准确、可靠,可为积雪草多指标成分质量评价提供参考。
Abstract
OBJECTIVE To establish a method for simultaneous determination of five typical triterpenes in Centellae Herba by a combinative method of HPLC fingerprint and quantitative analysis of multi-components by single marker (QAMS), and verify its applicability and accuracy in the quality control of Centellae Herba. METHODS The HPLC fingerprint analysis method for Centellae Herba was developed. Based on this, QAMS method was developed and evaluated by comparison of the quantitative results with the external standard method. RESULTS The HPLC fingerprints of Centellae Herba were established using 12 batches of samples. Twenty-three peaks were acquired as co-existing peaks and eight peaks among them were identified with chemical references. With asiaticoside as internal reference substance, QAMS was developed and the mean fs/k of asiaticoside B, madecassoside, madecassic acid and asiatic acid were 0.85, 1.13, 0.46 and 0.45, respectively. The contents of four triterpenes in 12 batches of Centellae Herba measured by QAMS method were not significantly different from those using the external standard method. CONCLUSION The established method is simple, accurate and reliable, which can be used for the quality evaluation of five triterpenes in Centellae Herba.
