Abstract��
OBJECTIVE To establish an UPLC-MS / MS method for simultaneous determination of a variety of drugs used for immunosuppressive therapy in the whole blood of renal transplant patients and to use this method for blood concentration monitoring in therapeutic drug monitoring. METHODS Blood of renal transplant patients was precipitated, and then cyclosporine A, tacrolimus, sirolimus, mycophenolic acid, prednisolone were measured by liquid chromatography-tandem mass spectrometry with two modes of ESI+ and ESI- in the multiple reaction monitoring (MRM) scan mode. Separation condition was gradient elution. RESUTLS This method of each drug had good linearity in the detection range. Accuracy, precision, matrix effects, methods of recovery and stability met the analysis requirements. Not only 970 plasma concentrations of 600 cases of TDM samples which has been routine determined but also additional 331 plasma concentration data were quantitatively analyzed by this method. XONCLUSION The method is simple, highly effective, and including comprehensive drug information, which applied to the blood concentration monitoring of multiple drugs in immunosuppressive treatment.
