Abstract��
OBJECTIVE To develop a method of quantitative analysis of multi-components by single marker (QAMS) for simultaneous determination of daidzein, daidzin, genistein and genistin in Semen Sojae Praeparatum. METHODS Chromatographic conditions were:Diamonsil-C18 column (4.6 mm��250 mm,5 ��m), mobile phase of methanol(A)-0.5% acetic acid(B),gradient elution (0-25 min,30%-45%A,25-50 min,45%-60%A),flow rate 1.0 mL��min-1,column temperature 30 ��,detection wavelength 260 nm. An HPLC method was developed as QAMS to determine the four components of soybean isoflavones in Semen Sojae Praeparatum. Daidzein was selected as the internal reference substance, and the relative correlation factors (RCF) of daidzin, genistein and genistin were determined by HPLC with good reproducibility. The contents of the four components in 15 batches of sample were determined by QAMS and external standard method. RESULTS No significant differences were observed between the quantitative analysis RESULTS of QAMS method and external standard method in 15 batches of Semen Sojae Praeparatum. CONCULSION It is feasible and suitable to evaluate the quality of Semen Sojae Praeparatum by QAMS.
