Abstract��
OBJECTIVE To investigate the effect of tetramethylpyrazine on extracellular DA,its metabolites and hydroxyl radical in striatum of PD rats treated by L-DOPA.METHODS The SD rats were randomly divided into control group,model group,high-dose tetramethylpyrazine group,low-dose tetramethylpyrazine group and GSH group.PD model rats were induced by intracerebral injection of 6-hydroxyl dopamine(6-OHDA).All rats in conscious and freely moving state were treated with L-DOPA using cerebral microdialysis technique.The change of extracellular DA,its metabolic productions and hydroxyl radical levels in striatum of PD rats were observed dynamically.RESULTS After being treated by L-DOPA,the DA concentrations of rats in control group were higher than those of rats in model group at 4 time points,the metabolism ratio of DA in model group were higher than those of control group at 9 time points,the concentrations of 2,3-DHBA and 2,5-DHBA(the index of hydroxyl radical) increased at 6 and 7 time points significantly(P<0.05 or P<0.01).Compared with model group,the lower level of 2,3-DHBA and 2,5-DHBA and metabolic ratio of DA at some time points were obtained in high and low dose tetramethylpyrazine group and GSH group(P<0.05 or P<0.01).CONCLUSION Tetramethylpyrazine can improve the DA metabolic ratio in striatum and reduce the oxidative damage in PD rats induced by L-DOPA.
