Abstract��
OBJECTIVE To study a new extraction method for analyzing diosgenin in Dioscorea nipponica Makino.---extraction in combination hydrolysis in situ.METHODS The effective factors on the yield of diosgenin were evaluated in order to establish the optimum conditions for extraction in combination hydrolysis in situ and the suitable method for isolating diosgenin from the crude extracts. RESULTS Hydrolyses was conducted in 1.5 mol��L-1 sulfuric acid with 70% isopropanol at 100�� for 4 h. The higher recovery of diosgenin was obtained by the extraction in combination hydrdysis in situ than that by traditional methods.CONCLUSION The method is simple, accurate and time saving.
