摘要

目的 建立分光光度法测定家兔血浆中硫酸化茯苓多糖的浓度。方法 将茯苓多糖硫酸化得到硫酸化茯苓多糖,利用光谱探针天青A与硫酸化茯苓多糖相互作用,其反应体系在625nm处吸光度与硫酸化茯苓多糖浓度成线性的特点,建立一种分光光度法测定家兔血浆中多糖浓度,线性方程A =1.135-7.397c,r=-0.998;线性范围0~0.4mg.mL-1。结果 灌胃给药后,在测定的时间段内(0.33~8h)硫酸化茯苓多糖在血浆中均有分布,给药1.5h时达最大。结论 本方法简单、稳定、可靠,可用于多糖血药浓度分析及其药动学研究。

Abstract

OBJECTIVE To develop a spectro photometric method for the determination of sulfation pachymaran in rabbit plasma.METHODS Pachymaran was sulfated into sulfation, pachymaran.The spectroscopi probe Azur A(AA)was interacted with sulfation pachymaran (SP).The absorbance decrease at 625 nm was in proportion to SP concentration.RESULTS The linear quotion of SP concentration and absorption value at 625 nm was:A=1.135-7.397 c,r=-0.998. The linear range was 0~0.4 mg·mL-1. After the sulfation pachymaran was given to the rabbit by ig,the sulfation pachymaran was found in rabbit plasma from 0.33h to 8h,and the peak concentration was reached at 1.5h.CONCLUSION Being simple, stable and credible,the spectro photometric method was able to determine the concentration of polysaccharides and their metabolism in rabbit plasma.
