摘要
目的:建立HPLC法测定家兔血浆中非诺贝特酸的血药浓度。方法:以吲哚美辛为内标,曱醇-0.02 mol·L-1醋酸盐 缓冲液PH 3.6(63:37)为流动相,检测波长为296 nm。结果:低、中、高浓度的日内回收率分别为97.02% ,97.66% , 101.17% ,RSD分别为1.41%,4.27%,1.01%;日间回收率分别为96.32%,93.51%,95.60%,RSD分别为 4.91%,4.70%,3.46%。结论:该法操作简便,结果可靠,适用于非诺贝特酸家兔血药浓度的测定。
Abstract
OBJECTIVE To establish a method for determination of fenofibric acid in rabbits plasma. METHODS Methanol-0.02mol·L-1 acetate salt buffer pH 3.6 (63:37) as mobile phase, indomethacin as internal standard,the wavelength of detection was 296nm. RESULTS The retention time of fenofibric acid and indomethacin was 3.80 and 6.56 min respectively. The intraday mean recovery of three different concentration of fenofibric acid were 97.02% (RSD=1.41%), 97.66% (RSD=4.27%) and 101.17 (RSD=1.01%)respectively, while the interday average recovery were 96.32% (RSD=4.91%),93.51 % (RSD=4.70%) and 95.60% (RSD =3.46%) in turn. CONCLUSIONS This method is sensitive with constant result and rapid for determining fenofibric acid concentration in rabbits plasma.
