目的:研究神经生长因子(nerve growth factor,简称NGF)在小鼠体内的药代动力学。方法:〈sup〉125〈/sup〉I-NGF用氯胺T法制备经静注和肌注两种途径,血装中NGF的浓度采用同位素示踪结合三氯醋酸(TCA)沉淀法或SDA-PAGE电泳法,测定小鼠血浆NGF的浓度。结果:静注〈sup〉125〈/sup〉I-NGF 25 μg·kg〈sup〉-1〈/sup〉,电泳法测得消除半衰期t〈sub〉1/2β〈/sub〉为2.223 h,酸沉法为2.264 h。酸沉法为2.264 h。肌注〈sup〉125〈/sup〉I-NGF 75,25,10μg·kg〈sup〉-1〈/sup〉电泳法测得消除半衰期t〈sub〉1/2β〈/sub〉分别为2.140,2.331,3.173 h,酸沉法测得消除半衰期t分别为2.480,2.552,2.642 h,达峰时间t〈sub〉max〈/sub〉为0.583 h,电泳法和酸沉法测得〈sup〉125〈/sup〉I-NGF平均血装清除率(Cl)分别为0.321和0.332 L·h〈sup〉-1〈/sup〉·kg〈sup〉-1〈/sup〉表观分布容积(Vd)为1.259和1.315 L·kg〈sup〉-1〈/sup〉,体内平均滞留时间(MRT)为3.592和3.539 h。结论:NGF在体内消除半衰期较短。本研究为今后的临床应用提供了必要的参考资料。
Abstract
OBJECTIVE:To study the pharmacokinetics of nerve growth factor (NGF) in mice. METHODS:125I-NGF was prepared by ChloramineT method.Its concentration in plasma was determined by trichloroacetic acid (TCA) precipitation method and SDSPAGE method after iv and im 125 INGF,respectively. RESULTS:After iv 125 I-NGF 25 μg·kg-1,the elimination half life (t1/2β) was 2.223 h in SDS-PAGE method and 2.264 h in TCA method,respectively.After im 125I-NGF 75,25 and 10 μg·kg-1,the elimination half life (t1/2β) was 2.140,2.331 and 3.173 h in SDSPAGE method,and was 2.480,2.552,2.642 h in TCA method,respectively.The time to peak (tmax) was 0.583 h in the two methods.By the use of the two methods,the mean plasma clearance (Cl) was 0.321 and 0.332 L·h-1·kg-1,the apparant volume of distribution (Vd) was 1.259 and 1.315 L·kg-1and the mean residence time (MRT) in mice was 3.592 and 3.539 h,respectively.
