目的 研究乙酰胡椒乙胺(PAA)对小鼠化学中毒性肝损伤的保护作用。方法 小鼠ig PAA后ip四氯化碳(CCl4)或D-氨基半乳糖(D-Gal),检测肝损害的几项指标和肝脏的病理损害。结果 PAA可明显降低CCl4,D-Gal引起的ALT升高,防止肝糖原下降,明显抑制由CCl4体外引起的大鼠肝微粒体脂质过氧化。结论 PAA对CCl4及D-Gal引起的肝损伤有明显保护作用。
Abstract
OBJECTIVE To study the protective effects of piperoethyl acetyl amine (PAA) on hepatotoxin induced liver injury in mice. METHODS Those mice were treated with a dose of PAA 1 h before or two doses of PAA 24 h before in injection of CCl4or D-galactosamine.Serum transaminase (ALT) and liver glycogen were determined.The damage of liver tissues was observed by light microscope.Malondialdehyde (MDA) production of liver microsomes initiated by CCl4in vitro was measured. RESULTS Administration of PAA prior to CCl 4 significantly prevented the elevation of serum ALT and the decrease of liver glycogen in CCl4 or D-galactosamine intoxicated mice.The damage of liver tissues was also reduced.CCl4initiated lipid peroxidation (MDA formation) of liver microsomes in vitro was inhibited by PAA in a dose-dependent manner. CONCLUSION The hepatoprotective action of PAA against CCl4and D-galactosamine intoxication in mice may be related to its inhibitory effect on CCl4initiated microsomal lipid peroxidation and the increase of liver glycogen.
