摘要

目的建立应用RP-HPLC同时测定灵芝中6种主要活性成分灵芝酸A、灵芝酸B、灵芝酸C2、灵芝酸G、灵芝酸E、赤芝酸A含量的方法。方法采用Diamonsil-C18色谱柱(4.6mm×250mm,5μm),流动相为乙腈和0.8%醋酸水溶液,梯度洗脱,流速1mL·min-1,检测波长254nm,柱温为室温。结果灵芝酸A、灵芝酸B、灵芝酸C2、灵芝酸G、灵芝酸E、赤芝酸A的质量分别在0.650~7.800,0.175~2.100,0.206~2.475,0.188~2.250,0.275~3.300,0.120~1.440μg的对数值与相应峰面积的对数值呈良好线性关系。平均回收率(n=6)分别为100.06%(RSD=2.6%),99.55%(RSD=1.6%),100.75%(RSD=1.5%),98.86%(RSD=1.9%),99.26%(RSD=2.3%)和98.95%(RSD=1.3%)。结论本方法简单、快速、准确可靠,可作为灵芝药材的质量控制方法。

Abstract

OBJECTIVE To establish a RP-HPLC method for simultaneous determination of six kinds of triterpenoid acids in Ganoderma,i.e.ganoderic acid A,ganoderic acid B,ganoderic acid C2,ganoderic acid G,ganoderic acid E and lucidenic acid A.METHODS The separation was performed on a Diamonsil-C18 column(4.6 mm×250 mm,5 μm),using a gradient elution with acetonitrile-wate(rcontaining 0.8% acetic acid)as the mobile phase.The flow rate was 1 mL·min-1,the detection wavelength was at 254 nm and the temperature of column was at room temperature.RESULTS The linear relationship were obtained within the range of 0.650-7.800,0.175-2.100,0.206-2.475,0.188-2.250,0.275-3.300,0.120-1.440 μg for the six components respectively.The average recoveries of the six components(n=6) were 100.06%(RSD=2.6%),99.55 %(RSD=1.6 %),100.75%(RSD=1.5 %),98.86 %(RSD=1.9 %),99.26%(RSD=2.3%) and 98.95%(RSD=1.3%).CONCLUSION The simple,rapid and reliable method could be used for the quality control of Ganoderma.
