Abstract:OBJECTIVE To optimize the extraction process of total flavonoids from Hedyotis diffusa, which have good anti-tumor effect, and improve the yield by combining enzyme extraction method and ultrasonic extraction method. METHODS Based on the total flavonoids content, the effects of enzyme type, ethanol concentration, enzyme dosage, enzymatic hydrolysis time and solid-liquid ratio on the extraction rate of total flavonoids from Hedyotis diffusa were studied by single factor experiment. Central composite experimental design (CCD) and response surface methodology (RSM) were used to optimize the extraction process. RESULTS When cellulose-pectin-wood protease was used, the ethanol concentration was 60%, the enzyme amount was 0.7%, the enzymatic hydrolysis time was 90 min, and the ratio of material to liquid was 1∶25(g·mL-1). The extraction rate of total flavonoids from Hedyotis diffusa was 1.95%. CONCLUSION The optimized extraction process of total flavonoids from Hedyotis diffusa by response surface methodology is reasonable and feasible, and the operation is simple. It can be used as a new method for extracting total flavonoids from Hedyotis diffusa.
