OBJECTIVE To develop the accelerated release test method of thienorphine hydrochloride PLGA micropheres. METHODS The accelerated release condition of thienorphine hydrochloride PLGA microspheres was established by examining the effects of temperature (37 ℃ and 55 ℃ and the concentration of hydrochloric acid in media release (0.000 01, 0.000 1, 0.01, 0.1 and 0.5 mol·L-1 on thienorphine hydrochloride released from microspheres. The regression equation was built by correlation evaluation of accelerated release data and long term release data. RESULTS Accelerated release rate correlated well with long release rate (r=0.977 2. CONCLUSION Accelerated release test could be applied to evaluate the drug release profile of thienorphine hydrochloride PLGA microspheres.
