摘要

目的 探讨灯盏花素对抗结核药致小鼠肝损伤的保护作用及其机制。方法 40只小鼠随机分为对照组、模型组、护肝片组、灯盏花素高和低剂量共5组。除对照组外,其余各组每天采用异烟肼和利福平灌胃造成肝损伤模型,2 h后,对照组和模型组分别予以等体积生理盐水灌胃,其余各组分别予以护肝片、灯盏花素灌胃,连续2周。分别检测各组小鼠肝指数和血清谷丙转氨酶(ALT)的活性;肝组织匀浆丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性及肝组织病理学变化。结果 灯盏花素和护肝片组均可明显降低肝损伤小鼠肝指数、血清ALT活性、肝匀浆MDA的含量,并能显著提高肝匀浆SOD的活性。肝组织病理学检查,灯盏花素组明显减轻肝细胞变性和坏死。结论 灯盏花素对抗结核药致小鼠肝损伤具有明显的保护作用,其机制可能与其抗氧化作用有关。

Abstract

OBJECTIVE To investigate the protective effect of breviscapine on hepatic injury induced by anti-tubercular drugs in mice and its mechanism. METHODS Forty rats were randomly divided into 5 groups (control,model,huganpian,high and low breviscapine). Oral isoniazid and rifampicin were simultaneously administered in mice except control group.After 2 h,normal saline was administered in control and model groups,huganpian and breviscapine were administered respectively in other groups once a day for 2 weeks. The liver index,level of alanine transaminase (ALT) in serum,content of MDA,activity of SOD in liver homogenate,and hepatic morphology were determined. RESULTS Breviscapine and huganpian significantly decreased the liver index,level of ALT in serum and content of MDA and increased the activity of SOD.Pathology examination showed that breviscapine evidently alleviated the degeneration and necrosis of hepatic cells.CONCLUSION Breviscapine has a protective effect on hepatic injury induced by anti-tubercular drugs in mice.One of the mechanisms of hepatic protection may be related to the antioxidative function.
