摘要

目的 建立同时测定独活中蛇床子素和二氢欧山芹醇当归酸酯含量的反相高效液相色谱法。方法 Shiseido C18 MG(4.6 mm×250 mm,5 μm)色谱柱,流动相乙腈-水(49∶51),检测波长330 nm,流速1.0 mL·min-1,柱温30 ℃。结果 蛇床子素在0.008 2~0.656 mg·mL-1内呈良好线性,r=1.000 0,平均回收率99.6%(RSD=2%);二氢欧山芹醇当归酸酯在0.003 6~1.536 mg·mL-1内呈良好线性,r=0.999 9,平均回收率99.4%(RSD=2.2%)。结论 通过对22批不同来源独活药材与饮片的含量测定,证明该方法简单,准确可靠,重复性好。在重庆巫山地区以海拔1 600~1 700 m、向阳、黑土的生长条件下,独活中蛇床子素和二氢欧山芹醇当归酸酯的含量均较高。

Abstract

OBJECTIVE To develop a quantitative reversed phase high performance liquid chromatography method for simultaneous determination of osthole and columbianadin in Radix angelicae pubescentis. METHODS The samples were separated at 30 ℃ on a Shiseido C18 MG (4.6 mm×250 mm,5 μm) column eluted with acetonitrile-water (49∶51) as the mobile phase. Flow rate was set at 1.0 mL·min-1 and the detection wavelength was set at 330 nm. RESULTS The calibration curve of osthole was linear within the range from 0.008 2 to 0.656 mg·mL-1 (r=1.000 0) and the average recovery was 99.6% (RSD=2.0%). The calibration curve of columbianadin was linear within the range from 0.003 6 to 1.536 mg·mL-1 (r=0. 999 9) and the average recovery was 99.4% (RSD=2.2%). CONCLUSION The developed method was proved to be simple, reliable and useful by analysis of 22 batches of the samples purchased from different areas. The samples collected in 1 600~1 700 miter altitude with abundant light supply and black soil accumulate much higher amount of osthole and columbianadin than those of other growth conditions Mountain Wu.
