目的 建立同时测定十味活血丸中苦杏仁苷和羟基红花黄色素 A 含量的 RP-HPLC 法。 方法 以十味活血丸为研究对象,采用 Diamonsil C18 柱 ( 4.6 mm × 200 mm , 5 μm) 色谱柱 , 流动相为甲醇 -0.05% 磷酸 (26 ∶ 74) ,流速 1.0 mL·min-1 ,检测波长 210 nm ,柱温 35 ℃ 。 结果 苦杏仁苷和羟基红花黄色素 A 质量 浓度分别在 2.40~ 24.0 g ·L-1 ( r = 0.999 8 , n =6) , 1.76~ 17.6 g ·L-1 ( r = 0.999 7 , n =6) 内与峰面积呈良好线性关系;精密度实验 RSD 分别为 1.1% , 0.59% ;重复性实验 RSD 分别为 0.74% , 0.89% ;平均加样回收率 ( n =9 ) 分别为 99.9%(RSD=1.2%) 和 99.1%(RSD=1.5%) 。 结论 本测定方法简便、快捷、准确,为 十味活血丸 质量评价提供依据。
Abstract
ABSTRACT:OBJECTIVE To develop a RP-HPLC method for simultaneous determination of amygdalin and hydroxysafflor in Shiweihuoxue pills. METHODS The separation was performed on a Diamonsil C18 (4.6 mm×200 mm, 5 mm) column with the mobile phase consisting of methanol-0.05% phosphonic acid (26∶74) at a flow rate of 1.0 mL·min-1. The UV detection wavelength was at 210 nm, and the column temperature was at 35 ℃. RESULTS The linear range was 2.40-24.0 g·L-1 (r= 0.999 8, n=6) for amygdalin, 1.76~17.6 g·L-1 (r=0.999 7, n=6) for hydroxysafflor. The average recoveries (n=9) of amygdalin and hydroxysafflor were 99.9%(RSD=1.2%) and 99.1%(RSD=1.5%), respectively. CONCLUSION The method is found to be simple, accurate and reliable for simultaneous analysis of amygdalin and hydroxysafflor in Shiweihuoxue pills.
