摘要

目的研究甲磺酸帕珠沙星对兔体内氨茶碱药动学的影响。方法采用自身对照法,以HPLC测定兔单用氨茶碱及与甲磺酸帕珠沙星合用后的茶碱血清浓度,计算药动学参数。结果经BAPP2.3药动程序拟合,甲磺酸帕珠沙星使兔血清中茶碱的曲线下面积AUC由152.0mg·h·L-1增加到361.4mg·h·L-1;半衰期t1/2由2.292h延长至6.823h;MRT由4.944h延长到9.383h,均有非常显著性差异(P<0.01)。结论家兔体内合用甲磺酸帕珠沙星对氨茶碱的消除药动学有显著抑制作用。

Abstract

OBJECTIVE To study the influence of pazufloxacin mesylate on the pharmacokinetics of aminophylline in rabbits. METHODS With a self-controlled method, the concentration of aminophylline was determined by HPLC in rabbits with aminophylline alone and in combination with pazufloxacin mesylate, and the pharmacokinetic parameters were also calculated respectively. RESULTS The mean plasma concentration-time profile of aminophylline was processed by BAPP2.3 pharmacokinetic software. The main parameters between single and combination indicated significant difference (P<0.01). The main parameters were as follows: AUC was 152.0 mg·h·L-1 and 361.4 mg·h·L-1; t1/2 was 2.292 h and 6.823 h;MRT was 4.944 h and 9.383 h, respectively. CONCLUSION Pazufloxacin mesylate can decrease the elimination of aminophylline in rabbits.
