摘要
目的探索中药配伍对中药复方药效成分药动学的影响。方法采用反相高效液相色谱法测定延胡索乙素的血药浓度,非房室模型拟合药动学参数,统计学处理采用t检验法。结果延胡索提取物组和延胡索白芷提取物组的主要药动学参数:ρmax分别为(0.484±0.30)和(0.853±0.52)mg·L-1,AUC0-∞分别为(1.822±1.0)和(5.542±3.8)mg·h·L-1,tmax分别为(1.06±0.83)和(1.88±1.6)h,两组药动学参数的差异具有显著性统计学意义(P<0.05)。结论延胡索与白芷配伍对延胡索乙素的药动学特征有显著影响,提示该配伍可能是导致延胡索乙素的药动学特征变化的原因。
Abstract
OBJECTIVE To investigate the influence of Chinese herb prescription Yanhusuo-Baizhi on pharmacokinetic properties of tetrahydropalmatine(THP) in dogs.METHODS The concentrations of tetrahydropalmatine in dog plasma were determined by HPLC method.The pharmacokinetic parameters were calculated using non-compartmental model and t test was used for statistical analysis.RESULTS The mean ρmax,AUC0~∞ and tmax of THP administrated with Yanhusuo-Baizhi extract were(0.853±0.52)mg·L-1,(5.542±3.8)mg·h·L-1 and(1.88±1.6)h,significantly higher(P<0.05) than those administrated with Rhizoma Corydalis extract.CONCLUSION There was significant influence of Yanhusuo-Baizhi prescription on pharmacokinetics of THP in dogs,suggesting the compatibility in drug combination of Yanhusuo-Baizhi.
