Determination of Adenosine in Different Parts of Acanthopanax giraldii Harms of Tibetan Drug in Sichuan by RP-HPLC

摘要

目的建立藏药红毛五加药材中腺苷的含量测定方法并比较红毛五加不同药用部位的腺苷含量。方法考察了回流法和超声法的优劣以及提取溶媒、提取次数、超声时间、料液比对腺苷提取率的影响;采用Kromasil C18(4.6mm×250mm,5μm)色谱柱,以乙腈-水(5:95)为流动相,流速1.0mL·min-1;柱温25℃;检测波长为258nm。结果腺苷的提取方法为以10倍量的水超声处理40min,提取1次;腺苷在0.07~0.69μg内呈良好的线性关系(r=0.9993),平均加样回收率为98.84%,RSD=0.80%(n=5);红毛五加各药用部位中的腺苷含量分别为:根0.47%,茎皮1.49%,茎刺0.42%,叶未检出。结论本方法简便、准确,重现性好,可用于红毛五加的质量控制,为该药材进一步开发利用提供科学依据。

Abstract

OBJECTIVE To establish a RP-HPLC method for the determination of adenosine in Acanthopanax giraldii Harms of Tibetan herbal medicine in Sichuan,and to investigate contents of adenosine in Acanthopanax giraldii Harms from different parts.METHODS The reflux and ultra sonic wave method were used.And the factors affecting the extraction of adenosine,such as the extracting solvent,the extracting times,the number of extraction and the ratio of solid to liquid were studied experimentally.The separation was performed on a Kromasil C18(4.6 mm×250 mm,5 μm) column with the mobile phase consisting of actonitrile-water(5∶95).The flow rate was 1.0 mL·min-1 and UV wavelength was at 258 nm.The column temperature was 25 ℃.RESULTS The optional conditions of extraction of adenosine were as follows:extracted by ultra sonic wave for 40 min with 10 times water.A good linearity was obtained in the range of 0.07~0.69 μg(r=0.999 3) with average recovery of 98.84% and RSD of 0.80%(n=5).The contents of adenosine in the root,bark and aculeus were 0.47%,1.49% and 0.42%,respectively,and the content was not determined in the leaves.CONCLUSION The method is simple,accurate and can be used to control the quality of Acanthopanax giraldii Harms,to provide scientific basis for future research.
