摘要
目的制备葛根素亚微乳,并对葛根素亚微乳体内的抗溶血进行评价。方法以复合物-相转变-高压乳化法制备葛根素亚微乳,以红细胞数,血浆结合珠蛋白,血浆血红蛋白,红细胞渗透脆性等血液参数值为评价指标,以市售葛根素注射液为对照,进行葛根素亚微乳体内抗溶血评价研究。结果相转变乳化法制备的葛根素亚微乳的理化性质稳定,平均粒径188.14nm,Zeta电位-29.03 mV,包封率89.72%。体内溶血实验结果表明,葛根素亚微乳组家兔的红细胞数,血浆结合珠蛋白,血浆血红蛋白,红细胞渗透脆性等水平与空白对照组没有显著性差异,而市售葛根素注射液组家兔出现了血管内溶血现象。结论相转变乳化法制备的葛根素亚微乳理化性质稳定,凭借亚微乳载药系统对葛根素的包合作用,减轻了葛根素血管内溶血的不良反应,更安全有效。
Abstract
OBJECTIVE To prepare puerarin submicron emulsions,and evaluate its anti-hemolysis side effect.METHODS The puerarin submicron emulsion was prepared by the novel complex-phase inversion-high press homogenization technology,and compared with puerarin injection,the protective mechanism of its anti-hemolysis activity was studied in terms of red blood cell values,packed cell volume,plasma hemoglobin level,haptoglobin level and osmotic fragility.RESULTS The mean diameter,Zeta potential and entrapment efficiency of emulsion prepared by this method were 188.14 nm,-29.03 mV and 89.72%,respectively.And the red blood cell values,packed cell volume,plasma hemoglobin level,haptoglobin level and osmotic fragility of puerarin submicron emulsion were significantly different(P<0.05),compared with those of control group.Puerarin submicron emulsion reduced the hemolysis activity of puerarin,but puerarin injection resulted in the hemolysis side effect in vivo.CONCLUSION Puerarin submicron emulsion prepared by complex-phase inversion-high press homogenization technology reduced the hemolysis side effect of puerarin by the incorporation of drug.Submicron emulsion is more safe and effective system for puerarin.
